質譜分析技術的原理是化合物分子經高能電子流離子化,生成分子離子和碎片離子,然后利用電磁學原理使離子按不同質荷比分離并記錄各種離子強度,得到一幅質譜圖。每種化合物都具有像指紋一樣的獨特質譜圖,將被測物的質譜圖與已知物的質譜圖對照,就可對被測物進行定性、定量。隨著信息化技術的進步以及色譜-質譜儀器分辨率和靈敏度等性能的不斷提高,只需要納克級甚至皮克級樣品,就可得到滿意的質譜圖。高分辨質譜測定的分子量精度可以達到百萬分之五(m/z可精確到小數點后第4位,即0.0001),加之質譜能提供化合物的元素組成以及官能團等結構信息,其對化合物定性、定量的準確度和靈敏度無與倫比。
關于食用農產品中農藥殘留檢測技術,龐國芳科研團隊檢索了近二十年(1991—2010)國際上有一定影響力的15種期刊SCI論文3505篇,涉及檢測技術200多種。對論文總量排名前20位的技術,按前十年(1991—2000)和后十年(2001—2010)發展歷程進行對比研究發現:前十年發表的色譜-質譜農藥殘留檢測技術論文有339篇,而到后十年達到了1018篇,后十年約是前十年的3倍,二者之和1357篇,約占總量的39%。過去二十年發展最耀眼的分析技術是LC-MS/MS和GC-MS/MS,其中,發展最快的技術是LC-MS/MS,它由前十年的第9位上升到后十年的第1位;GC-MS/MS由前十年的第19位上升至后十年的第8位。這充分說明,在食用農產品農藥殘留檢測技術方面,色譜-質譜檢測技術已迎來了空前發展的新時期。我國這一領域科技工作者緊跟這一技術的前進步伐,使我國由前十年的第14位,躍升到后十年的第2位,為我國在這一領域國際地位的提升做出了突出貢獻。
基于色譜-質譜聯用分析技術的獨特優勢,龐國芳科研團隊從2000年至今一直從事農藥殘留高通量色譜-質譜方法學研究,他們采用當前國際上農藥殘留分析領域普遍關注的先進技術,包括氣相色譜-質譜、氣相色譜-串聯質譜、氣相色譜-四極桿-飛行時間質譜、液相色譜-串聯質譜、液相色譜-四極桿-飛行時間質譜和線性離子阱-電場回旋共振軌道阱組合質譜共6類色譜-質譜聯用技術,評價了世界常用1300多種農藥化學污染物在不同條件下的質譜特征,采集了數萬幅質譜圖,形成了《世界常用農藥色譜-質譜圖集》,分五卷出版:第一卷為《液相色譜-串聯質譜圖集》,第二卷為《液相色譜-四極桿-飛行時間質譜圖集》,第三卷為《線性離子阱-電場回旋共振軌道阱組合質譜圖集》,第四卷為《氣相色譜-串聯質譜圖集》,第五卷為《氣相色譜-四極桿-飛行時間質譜及氣相色譜-質譜圖集》。這是一項色譜-質譜分析理論的基礎研究,是龐國芳科研團隊的原創性研究成果。他們站在了國際農藥殘留分析的前沿,解決了國家的需要,奠定了農藥殘留高通量檢測的理論基礎,在學術上具有創新性,在實踐中具有很高的應用價值。
根據這些質譜圖與建立的相關質譜數據庫,龐國芳科研團隊已經研究開發了水果、蔬菜、糧谷、茶葉、中草藥、食用菌(蘑菇)、動物組織、水產品、原奶及奶粉、蜂蜜、果汁和果酒等一系列食用農產品農藥殘留高通量檢測技術。同時,經過標準化研究,已建成20項國家標準,每項標準均可檢測400~500種農藥殘留,其操作像單殘留分析一樣簡單,卻比單殘留分析提高工效數百倍,在食品安全領域得到了廣泛應用。其中,茶葉農藥殘留高通量檢測技術2010年被國際AOAC(國際公職分析化學家聯合會)列為優先研究項目之一。經過4年準備,龐國芳科研團隊2013年組織了有美洲、歐洲和亞洲11個國家和地區的30個實驗室,共56個科研小組參加的國際AOAC協同研究。協同研究結果證明,各項指標均達到了AOAC技術標準,被推薦為AOAC官方方法,體現了這項研究的先進性和實用性。同時,也展示了我國學者在農藥殘留高通量檢測技術領域的水平和能力,擴大了我國在這一領域的國際影響,為世界農藥殘留分析技術的進步做出了突出貢獻。
